  • Although great effort has been made to specifically target tumour cells that express AR, the majority of individuals with prostate cancer experienced failure from these treatments.
  • Here, we use in vivo tests and mouse models to show that deletion of AR in stromal sonic-hedgehog Gli1-lineage cells reduces prostate epithelial oncogenesis and tumour formation.
  • Single-cell RNA sequencing and other analyses revealed a significant rise in insulin-like growth factor (IGF) binding protein 3 expression in AR-deficient stroma through attenuated AR (androgen)suppression on Sp1-regulated transcription.
  • This further inhibits IGF1-induced Wnt/-catenin activation in neighbouring basal epithelial cells and suppresses the growth and development of tumours in these cells. Mice lacking in stromal AR can regain IGF1-induced oncogenic growth in epithelial organoids.
