A luciferase assay acitivity is a reporter assay that is commonly used to make enquiry about expression of genes at the transcription. It is used to make enquiry  if a protein can activate or repress the expression of a target gene.

The data file attached below; shows the record of the  Analysis of the luciferase activity of T lymphocytes containing plasmids with different amounts of the CD3γ regulatory sequence.

We will see that the pCD3γ-59 contains the core promoter sequence as it possess the maximum effect on Luciferase expression. To calculate the increase in luciferase activity  ; we will need more data to perform more deletions for a confirmatory result in order to calculate the increase in luciferase activity
