We couldn't be where we are without microscopes and other technology. We couldn't study almost anything we know without these modern inventions.

They make tests and observations more accurate. Hundreds of years ago a theory wasn't as easy to prove as it is now, and even most of those that were "proven" to be true have been disproven in recent years.

While both will equate changes in a genetic chain, deletion would have more severe consequences.  A substitution mutation usually only causes minor disorders and diseases, like sickle cell anemia.  Whereas deletion causes much more serious diseases, like cystic fibrosis, and turner syndrome which is  where a female is born with only one X chromosome.
