These sentences connect to a previous one after a full stop by means of  conjunctive adverbs. Commas are required.  

Meanwhile, Cassie collected shells on the beach.  

I believe, therefore, that this is the best course of action.  

Not used correctly:

Instead should be placed at the end.

You should not sing in the concert if you have a cold, however.
