You perform a ligation with NcoI/NotI-digested pET-41a vector and the egfp insert from pEGFP-N1 (as in the lab). You then transf

orm the ligation mix into E. coli and plate on LB medium containing kanamycin. Consider the following unexpected outcomes, and suggest controls: a. Nothing grows. What controls might you design to determine whether the E. coli cells are viable (alive) versus whether the E. coli is competent

Explanation:

You can do two controls:

1) Check if the cells are viable by plating the transformed E.coli on LB medium with no antibiotics. If the cells are alive, a lawn will be seen, as there is no selection pressure. If the cells are dead, no colonies will be seen.

2) Check if the cells are competent by transforming them with a control plasmid. This is another plasmid, either commercial or already used in the lab, that contains the gene for resistance to kanamycin, and is known to work. If this positive control is used to transform the cells and still no growth is seen in the LB plates, then the E.coli are probably not competent.

postnew [5]

Answer:

light is required to provide energy for photosynthesis

Explanation:

For photosynthesis to take place, it requires the presence of light and chlorophyll. The process has two stages; light and dark stage.

In the light stage, chlorophyll molecules absorb light energy which is used to split water molecules into oxygen and hydrogen atoms. The process is called photolysis.

water→ hydrogen atoms + oxygen.

The hydrogen atoms produced enter the dark stage while oxygen is released in the atmosphere. In the dark stage, carbon dioxide combines with the hydrogen atoms to form simple sugar such as glucose.

Heart disease affects the cardiovascular system by plaque building up on the artery walls narrowing them making it harder for blood to flow through which could lead to heart attack or probably stroke if the plaque blocks it entirely or maybe almost entirely.

Heart disease may also affect our nervous system for there is higher risk to stroke and it is due to limited flow of blood to the brain and may lead to heart failure which may happen with heart disease that puts the stress on the brain and nerves.

The digestive system is affected by heart disease that causes the system to not get the right amount of oxygen and the nutrients that it needs from the blood.

What kind of bond is formed when one atom steals an electron from another atom
Delicious77 [7]
Ionic bonds are the type of bonds where there is transfer of electrons from one atom to another. The electrons are removed and from one atom and attached to another. A good example is salt which is composed of sodium and chlorine. Sodium readily loses one of its electrons and chlorine readily accepts it. Before losing the electron, sodium has a positive charge, but then becomes negatively charged after giving up the electron. Chlorine has a positive charge before gaining the electron but becomes negatively charged after gaining the electron. These opposite charges between sodium and chlorine attract the two elements together to form the ionic bond.
