The acidic lumen of the thylakoid will have greater concentration of H+ ions. when these ions are passed to basic pH of 8 then H+ ions tend to go to outside due to their concentration gradient, when is greater in lumen and lesser in the surrounding. the thylakoid holds ATP synthases that uses proton motive force H+ ions and will make the ATP. Hence an isolated chloroplast is capable of synthesizing ATP when the lumen is made acidic and the outer medium is made alkaline.
