The chromosome's attachment of spindle microtubules to the kinetochore, a complicated structure, is necessary for reliable chromosome segregation.

The abrupt division of the nuclear envelope into several tiny vesicles, which will later be divided between the future daughter cells, marks the start of prometaphase. Spindle assembly requires the breakdown of the nuclear membrane as a necessary step. Animal cells' centrosomes are outside the nucleus, thus until the nuclear membrane separates, the forming spindles microtubules cannot access the chromosomes.

During prometaphase, chromosomes are pulled and tugged in opposite directions by microtubules growing out from both poles of the spindle. Sister chromatids do not break apart during this tug-of-war because they are firmly attached to each other by the cohesin remaining at their centromeres. Chromosomes are bi-oriented at the conclusion of prometaphase, which means that the sister chromatids' kinetochores are attached by microtubules to the spindle's opposing poles.
