1. En la novela escrita por  Rómulo Gallegos Doña Barbara fue violada por  piratas que mataron a su novio, en la versión adaptada en 1943 eran trabajadores de sus padre los que le robaron, la violaron y mataron a su padre y novio.

2. En el libro Santos Luzardo regresa a su hacienda luego de estudiar derecho, en cambio en la novela Santos Luzardo regresa a vender su hacienda.

3. En el libro no se menciona que Doña Barbara mata a sus abusadores, pero en la adaptación de 1943 menciona que ella mata a sus cinco abusadores.

La novela literaria y las diferentes adaptaciones siempre presentan algunas diferencias por ejemplo en la película adaptada en 1943 las personas que le hacen daño a Doña Barbara son trabajadores de su padre, los cuales les roban y matan al padre y novio, además en la obra literaria Santos es un joven que regresa para ayudar a crecer la hacienda de su familia y en la película tiene intenciones diferentes. Es interesante que la adaptación de 1943 el escritor Rómulo  Gallegos, ayudo a escribir el libreto para la película.
