Gram staining is used to identify bacterial species which are either gram positive or gram negative.

The cell wall of gram positive bacteria contain thick layer of peptidoglycans which hold the stain crystal violet and thus it becomes purple color where as the cell wall of gram negative bacteria  contain thinner layer of peptidoglycans which allows the washing of crystal violet stain and thus the cell wall is not stained with purple color.
