Prokaryotes have single circular molecule of DNA which has only one origin of replication per circular chromosome.  

While Eukaryotes have several origin of replication. The chromosome of Eukaryotes is linear and long due to which it has multiple replicating sites with different replication timing.  With multiple replication, the process of transcription in eukaryotes becomes fast and easy.
