After cloning the Glo gene in the puc18 plasmid of E.Cori, the next step would be to culture the clone.  While amplifying DNA requires Polymerase Chain Reaction cycles, amplifying proteins requires the metabolic activity of <span>organisms to utilize central dogma process and ribosome activity to produce proteins.  Culturing produces many clones that synthesize the proteins in significant amounts. </span>
