DNA replication occur by a semiconservative model. During DNA replication the strands of DNA are unbounded by DNA helicase and stabilized by single cell binding proteins.

The primase enzyme build the short primers and DNA polymerase synthesize the new strand of DNA. One restriction for DNA polymerase is that it can only synthesize the DNA strand in 5' to 3' direction. That's why 5' end of DNA is always synthesized first.

The leading strand is synthesized continuously and lagging strand is synthesized in fragments known as okazaki fragments because of DNA polymerase ability to synthesize the strand in only 5' to 3' direction. These fragments are joined by DNA ligase and RNA primers are also replaced by DNA polymerase.
