Specialized cells are formed when cells differentiate - this is true. When you have stem cells, you're talking about undiferentiated cells with no specific roles in the body. When these cells start differentiating and assuming different roles, they are also becoming specialized.
In metaphase I, the homologous chromosomes line up at the metaphase plate. In anaphase I, the homologous chromosomes are pulled apart and move to opposite poles. Sister chromatids are not separated until meiosis II.
