It was "prominent slaveholders" who initially established the American Colonization Society, since this society was meant to strengthen the institution of slavery but separating free blacks from slaves.

The American Colonization Society was a group created in 1816 with the purpose of helping freed slaves to migrate to Africa, as it was believed in Africa  former slavers could have more opportunities than in the U.S. This idea was supported mainly by important slaveholders that believed this idea would help them to remove free slaves from the U.S. and by some religious groups that believed in the U.S. free blacks could not be as free as in their ancestors land in Africa. This led to the creation of Liberia in the west of Africa, a colony where thousands of freed slaves migrated; however, many of them died as a result of disease and other condition in Liberia and many more opposed the American Colonization Society because they wanted to state in the U.S. and believed this was just a strategy for continue with slavery in the U.S. and avoid freed slaves fought for equality. Considering this, it can be concluded the Amecian Colonization Society was mainly established by prominent slaveholders.
