The look in his eyes shows his drive to kill and he’s ready as well as killing the old woman shows he’s ruthless and cold blooded. The dressing of his arm shows he is prepared and used to these type of situations.
Curley mistakenly thinks that Lennie is laughing at him with the other men, when, in fact, Lennie is not aware of the drama between Curley and Slim and is simply chuckling to himself with glee thinking about the dream farm.
