In plants, carbon dioxide (CO2) enters the leaves through stomata, where it diffuses over short distances through intercellular spaces until it reaches the mesophyll cells. Once in the mesophyll cells, CO2 diffuses into the stroma of the chloroplast, the site of light-independent reactions of photosynthesis.

This media have phenol red as an indicator. When the glucose is fermented to acid , the production of acid turn the indicator from red to yellow, but it is then reoxidised and turns red again . When lactose is fermented it produce large amount of acid , and turn indicator yellow . Hence the slant will become yellow.And combination of ferrous sulphate and sodium thiosulphate helps in detection of H₂S which produce black color at the butt .

A large surface area plays a major factor for diffusion and also osmosis at some point. The larger the surface area, the more molecules are able to diffuse in and out of the cells. Think of it this way. You have 2 containers. One container have a length of 5cm by 5cm. Another container you have a 10cm by 5 cm. You have a bucket of water. Por the water into the container, it would be more easier to pour it into the 10 by 5 without spilling right. Thats because it has a larger surface area compared to the other container. Hence, the size and shape affects the influence of diffusion.
