Let's say that the story is set in a dark and spooky forest, the story will then have a dark theme/tone, compared to say, a story set in a magical castle with rainbows and what not. The setting the story is set in can help depend on whether or not the story will be unfortunate, scary, or happy. You won't expect good things to happen in a dark spooky forest.
