President Wilson is referring to the fact that he wanted world peace, and to have world peace, no country would try to defeat another to increase their power or reputation. The significance of this statement is because he wishes that there wouldn't be another world war (which there was, World War II), and that he wished everybody could work together from then on

By saying that "“The day of conquest and aggrandizement is gone by" Wilson is saying that it is time for a new world order, one in which territorial subjugation is not routine. The significance of this statement is he wishes that there wouldn't be another world war, that we could achieve world peace and order, and that he wished everybody could work together from then on and overcome differences and disagreements .
