Hamlet had organised a performance of a play where the very deeds of a younger brother killing his elder brother for the kingship were shown. Gertrude wanted Hamlet to apologize to his step father/uncle, the now king Claudius. In this scene, Hamlet pleads with her to change her ways, reveal the truth and become the lady she was before she married Claudius. This scene also ended in the accidental death of Polonius, Ophelia's father.

It was Gertrude that he wanted and offered a chance to align with him. Ophelia was the woman he loved who turned insane after the death of her father.
