In indole-positive bacteria, Tryptophan is used to form indole. This reaction is catalyzed by a tryptophanase enzyme.

Tryptophanase or tryptophan indole-lyase is an enzyme that catalyzes the reversible beta-elimination of Tryptophan to form indole.

Tryptophanase is present in many Gram-negative bacteria.

The indole-positive bacteria include <em>Escherichia coli, Haemophilus influenzae, Aeromonas hydrophila, A. punctata, Bacillus alvei, Klebsiella oxytoca</em>, etc.

On the other hand, the indole test is a test used to determine the ability of bacteria to convert tryptophan into indole.
