The proteins are one of the most important components of the living body. These proteins are formed of the monomers called amino acids. These amino acids are polymerisation in a two dimensional structure by means of peptide bond. This is the bond between the amino group of the preceding group with the carboxylic group of the succeeding group. This structure is called the primary structure. The primary structure then undergoes several intermolecular bonding with the side groups via covalent bonds, hydrogen bonds, disulphide bonds etc which forms the secondary structure which is the 3d structure.

The active site of the enzymes are formed by a particular structure of amino acids which are complementary to that of the substrate. So with the alteration of that site, the enzyme cannot act any more. Thus the enzyme becomes non functional.
