During DNA replication Helicases are the enzymes which separates the two strands of DNA forming replication fork.

DNA polymerase add DNA nucleotides to form new strand. DNA polymerases does not add nucleotide itself instead need a small RNA fragment called primer which bind with the template DNA strand. Once primer bind the DNA polymerase add nucleotides to the new strand.
