The optimum temperature for the enzyme in the above graph is the human body temperature (37.5 degree Celsius for the enzymes in the human cells.) Denaturation takes place at above 40 degree Celsius.

Explanation:

With temperature enzyme activity increases which clearly says that they are directly proportional to each other. But at When the temperature becomes very high the enzyme gets denature, which also changes the shape of the enzyme. If the shape of the enzyme changes it becomes difficult to bind with the substrate. Denature is the breakdown of the enzymes, when temperature increases they breakdown because of breaking of intra- intermolecular bonds are broken down because they starts getting more kinetic energy.
