Light-dependent reactions form the NADPH when the electrons from the water molecule are passed from PS II to PS I and finally to the NADP+. The enzyme NADP reductase catalyzes the reduction of NADP+ into NADPH. The enzyme is located on the stromal side of the thylakoid membranes and therefore, the NADPH is released into the stroma. Stroma is the site for the reaction of the Calvin cycle. During the Calvin cycle, the reduction of 3-phosphoglycerate into glyceraldehyde 3-phosphate uses NADPH as the electron donor.
