Protease refers to a protein-digesting enzyme, which functions on particular pH, for example pepsin functions on pH 2 inside the stomach, that is, highly acidic.  

An enzyme's shape determines its activity as they exhibit active sites that possess specific configurations, which fit in the substrate and lead to chemical reactions. A prime modification in pH can change the configuration of the active locations of the enzyme, making it sedentary.

Negative feedback is a term used in association with homeostasis. It is the biological process used to antagonize a stimulus in order to return the body to homeostasis.

For example, the normal body temperature of humans ranges between 36.5 to 37 degree Celsius. Beyond this range, the normal body temperature has been breached.

If it is not returned to normal within a short period, such a person is at risk of suffering irreversible damage to their body or even death.

Thus, in order to return the body system to the normal body temperature, negative feedback will be activated. This feedback will work in the opposite direction to the process causing a breach of the normal temperature range.

By doing so, the body temperature will quickly return to normal.
