The volume of the cube would be: side^3= 100mm^3=1,000,000 mm3

If broken into 10mm cube, it would form 1000 cube. The total area of the cube would be: 1000 * 6* 10 * 10= 600,000 mm2

After the cloned gene is altered it can be re-introduced back into the wild-type  yeast. The yeast is then allowed to multiply and the effect that the mutation has on the yeas can be observed.
