The very first time we see the handkerchief is just after Othello starts suspecting his wife of being unfaithful.

The handkerchief was the first gift from Othello to Desdemona because it was her

first remembrance from the Moor in which

Cassio finds the handkerchief and takes it to his woman, Bianca and from Bianca to Lago in which Iago puts the handkerchief

in Cassio's home, and tells Othello,

Bianca later come to give the handkerchief back, angry because she believes "This is some minx's token." in Othello recognizes it as his handkerchief
