I originate from being mexican, my best guess for this question is that it is asking you to replace "la casa" with the choices of 1-6 below.

in english it says

"the house.

lupita, where is your house?"

which doesn't make sense so you would want to pick number 2. La hermana

so it is able to say  

"la hermana

Lupita, ?donde esta tu casa?"

translating to

"the sister,

lupita, where is your house?"
