A point mutation can be insignificant due to the genetic code, many amino acids come from similar codons (where one base can be incorrect and it results in the same amino acid). If it does change the amino acid, having only one different amino acid in a whole polypeptide chain usually does not cause a major problem.

However, a change in the reading frame can be a major issue because it changes every codon form what it should be. A frame shift mutation can even prevent the translation of a protein entirely if it effects the start or stop codon.
