As you can see in the picture presented below, the DNA sequence in individuals with normal hemoglobin and in the individuals with sickle cell hemoglobin differ in only one nucleotide. However small, this difference in the nucleotide changes  the amino acid from glutamate to valine, changing the structure of the protein in the red blood cells.
