b. They represent microscopic examination

c .Gram positives take the purple color Gram negatives take the pink color.

d. The reagent is acetone alcohol, because it causes shrinkage of the peptidoglycan layer.

<h3>What is differential staining?</h3>

Differential staining is a staining process which involves the use of more than one chemical stain.

The use of  multiple stains allows for better differentiation between different microorganisms or structures/cellular components of a single organism.

a. The nature of A and D might be their ionic properties and this could be as a result of high content of Iron.

b. Reagents in gram staining represent differential stains used for the microscopic examination of bacterial cultures and laboratory specimens.

c. The appearance of gram positive is purple and gram negative is pink

Gram positives take the color of the primary stain during gram staining which is crystal violet- purple

Gram positive take the color of the secondary stain which is safranin- pink

d. The colorless reagent which is the acetone alcohol should not exceed the recommended time because it would dehydrate and shrink the peptidoglycan layer of the bacteria leading to a false result.
