Exosomes are small vesicles of size 40-90 nm. They are released by antigen presenting cells.  According to recent research, exosomes have shown to play a role in cell-cell communication.

Exosomes have a high degree of MHC Class II expression because they come from MHC class II peptide loading compartments. Three distinct and highly polymorphic isotypes, HLA-DR, -DQ, and -DP, respectively, encode the human MHC class II.

For the purpose of precisely isolating exosomes from cell-free supernatants, magnetic beads have been coated with monoclonal antibodies specific for HLA DP, DQ, and DR.Different molecules can be purified using magnetic beads, which is an easy and dependable technique.

Exosome-coated beads are then stained with conjugated antibodies and subjected to flow cytometric analysis. This approach can characterize exosomes revealing MHC Class I and II molecules and their associated cells.
