Chinese  hamster ovary cell production of recombinant tissue‐type plasminogen activator  (t‐PA) was increased by amplification of cotransfected dihydrofolate reeducates cDNA using stepwise adaptation to increasing methotrexate (MTX) concentrations. The highest producing clones were isolated at 5 μM MTX and yielded 26,000 U/106 cells/day t‐PA (43 μg/106 cells/day). Above 25 μM MTX, cell specific t‐PA production rates became increasingly variable and the cDNA copynumbers decreased. No apparent correlation between the cell specific t‐PA production rate and the growth rate was observed upon sub cloning of the amplified cells. When MTX selection was removed, the t‐PA production rate decreased up to tenfold within 40 days; this was accompanied by an up to 60% drop in cDNA copynumber. Subclones isolated after 108 days of culture in the absence of MTX were, on average, sixfold more stable than their parental cells. In culture without MTX, the maximum stable t‐PA production rate obtained (over 250 days) was 7000 ± 750 U/106cells/day (∼12 μg/106 cells/day), approximately threefold lower than the maximum unstable levels of production reached under selective pressure. Taken together, these results define a wide range of the highest t‐PA expression rates obtained under MTX selection, for which stable expression without selection has not been reported

medical assistants to ensure the personal and physical safety of stroke and seizure patients during an office visit.
musickatia [10]

In general medical assistants note the history and important signs of patients and maintain the record, to solve and understand healthcare problems.

Medical assistance will be done in hospitals, physicians chambers, RMP guest house and other medical facilities.

Medical assistant make sure that the personal and physical safety or hygiene of stroke and seizure patients whoever visited to the healthcare clinic. If there is no proper physical care the stroke patient mentally gets disturbed, leads to increased stroke may led to life threatening.

Taking personal care is very important for seizure patient, first aid will be given to the patients until seizure stops then prescribing anti-seizure medication is essential to maintain the critical health condition of the patients.

How can reduced precipitation lead to limited water availability
umka21 [38]
Reduced precipitation can lead to limited water availability by reducing the amount of rain that we get therefore there is less water. Without the rain the earth would eventually dry out and there would be no more water to use.

Is selection more effective against recessive alleles in haploid or diploid organisms?
svet-max [94.6K]
The selection of more effective which is against recessive alleles in haploid organisms than diploid organisms. This is because haploid organisms contain a single set of alleles if a deleterious allele is present in haploid organisms which will produce its effect immediately as there will be no dominant allele which can prevent the expression of the recessive allele as it happens in diploids. Recessive allele will not produce its effect in presence of the dominant allele in the case of heterozygote which is Aa.
Where there is haploid the selection will be more effective when removing recessive alleles in the population. It is the homozygous recessive condition which as aa then the selection will act against recessive alleles.
