The major difference that resulted in political parties was over how much power the Federal Government should have.  the founding fathers were against the formation of poliitical parties. They believed that it would divide the nation causing different groups or parties to fight for their own interest and not for the best interest of the nation. As time went own different leaders viewed the constitution in different ways with Hamilton believing the nation should take a broad view of the constitution with the national government having more power.  On the other hand, Jefferson was in favor of more of a narrow view with the power being in the hands of the people and the individual states.

Hamilton introduced the idea of a government bank. The bank would collect taxes, make loans and issue paper money. Jefferson was againist the government bank.

Jefferson sought a strong relationship with Frane while Hamilton did not

Manifest destiny was that the americans were destined to expand out towards the west and transportation/railroads and stuff allowed the easier access out to the west to start towns/new lives while being able to have some benefits of civilizations. You know, that kind of stuff. 
Manifest destiny is that it is inevitable and destined for americans to spread across the rest of the US continent.
