Acid fast staining is a staining technique that is used to differentiate two types of gram positive cells, those that have waxy mycolic acid in their cell wall and those that do not. 
Those cells that do not have mycolic acid in their cell wall will show blue color after acid fast staining.<span />

The acid-fast organisms such as mycobacteria can be differentiated from the other bacteria by the help of acid-fast staining. These acid-fast bacteria show a very high concentration of mycolic acid in the cell walls. The two stations that are used in this type of staining technique are carbolfuchsin and methylene blue.

The acid-fast organisms retain the red color of carbolfuchsin even after the decolorizing agent is added to it. The non-acid-fast organisms do not retain the red color of carbon fusion and thus, are counterstained by methylene blue.

All the gram-positive bacteria are acid-fast bacteria. In addition to the peptidoglycan layer, the acid-fast bacteria have a cell wall containing large amounts of glycolipids (mycolic acids). <em>Mycobacterium tuberculosis and Mycobacterium leprae are the two examples of acid-fast bacteria.  </em>
