With serial dilution the concentration reduces with which the number of bacteria will also reduce that will make the counting easier, that means there are millions of bacteria present in the cell culture and we need to get down to 30 to 200 cells which makes the count easier. As we know the concentration of dilution we have created that will help in calculating the exact number of bacteria present in the cell culture after multiplying with dilution factor.
  • If the population of the bacteria in the sample is too high, it will make it prone for the agar plate to lawn colonies to cover the whole agar plate and we cannot even count any of it.
  • The easiest selections to understand the improvement in the growth of bacteria is when maximum performance is needed either by exogenous or genetic means. So in those cases, full growth curve of colonies may not be needed.

1. It can be hard to count undiluted populations as there are so many, by diluting, the count can be done simpler and verified by the average of different counts. 

2. Bacteria colonies are likely to cover the entire plate if the number is high, therefore making it virtually impossible to count them.
