Lysosomal enzymes are produced in the endoplasmic reticulum (ER), and then they are exported to the Golgi complex, where mannose-6-phosphate (M6P) label is added to be finally packaged into acidified vesicles. Mutations in the genes that encode for lysosomal enzymes are known to produce Lysosomal Storage Diseases (LSD) including Tay-Sachs disease.
