It binds to and dissolves the hydrogen bonds connecting the bases on the DNA strands. These hydrogen bonds continue to be broken as the helicase travels along the DNA molecule, separating the two polynucleotide chains.

Primase, a different enzyme, momentarily binds to each strand when the helicase separates them in order to build a foundation upon which replication can start.
