Scientists used the mitochondrial DNA of Prince Philip in order to investigate the relationship of Tsarina with the Russian royal family.

Mitochondrial DNA inherited only through  the maternal line i.e., it is passed from the mother to her offspring.  

It helped in identifying the relationship of Tsarina with prince Philip who was related to Tsarina through his mother. It proved that Tsarina  and her four daughters were indeed buried into two graves.
