This is a rather personal question, and I can't answer for you, but I can give you my opinion.
When Macbeth says that line, he is referring to the meaninglessness of life. He is saying that life isn't really worth living because it is just a fleeting moment in time, and will eventually end, no matter what you do about it. He tried hard to fight for his ambition and become a king, and he did, but he died nevertheless, thus reaffirming his belief that life is 'signifying nothing' in the end.
Personally, I disagree with his opinions. Yes, life is going to end and we aren't going to live forever, however, we have to make do with what we were given. We aren't supposed just to lie down and wait to die - we should live our life to the fullest and make the most of it while we still can. As the famous aphorism goes, Carpe diem (Seize the day)!

I believe that people could be more tolerant if they have traveled more or have learned about their culture. If someone has traveled a lot, then they could understand the struggle of moving from one place to the other and dealing with difficult changes. Changes in time differences, weather, language, culture, and just people, in general, are all very difficult to go through, especially alone. If a traveler met someone from a different place, then they will most likely be very nice to them since they know what they had to go through to get to a new place. Being in the military would definitely make you more tolerable in that kind of situations since you move around a lot for the job. I also believe that being in a language studies class, or just learning about language and culture, could definitely help people become more open-minded. This goes a lot with travelers, you learn a lot about the people who speak the languages that you are learning. You then would be able to see how different their lives are in those kinds of places, and it would make you be more open-minded and caring to those people once you meet them, because then you know how much they might be struggling to adjust to their new lifestyle. It's difficult for some people to understand how hard it is to adjust to a new lifestyle or to move and travel in general, so learning about those things and taking classes for it really helps. I really do believe that they help you become more open-minded and tolerable to those kinds of people.
