-A scanning electron microscope (SEM) scans a focused electron beam over a surface to create an image. It useful for studying details of a specimen's surface.

-An electron beam passes over the specimen's surface and displaces electrons, which are then focused on a screen to form an image. Uses a conductive material (gold metal) on the specimen.

-The scanning electron microscope gives much higher clarity of a specimen being viewed than the light microscope can; therefore, internal structures of cells, such as organelles, DNA can be viewed in much more detail. Bacteria and viruses can also be viewed in details and studied under a scanning electron microscope.

Gram-positive bacteria have a thick peptidoglycan cell wall and dyes purple in the Gram test (for categorization of bacteria). Gram-negative bacteria have thin peptidoglycan layer and do not turn purple. Gram-positive bacteria are easier to kill because their thick peptidoglycan layer absorbs antibiotics.
