Colemanhypothesis states that ob+ genes codes for an appetite suppressing hormone. The amount of adipose tissue determines the production of this hormone. Also, db+ genes codes for receptor of the same hormone. So, mutations in these genes might have the following effects:When ob genes are mutated, production of functional appetite suppressing hormone stops.When db genes are mutated, production of functional receptors for the same hormone stops.

So, if the mutation occurs in both the ob genes, the production of appetite suppressing hormone stops and thus, the mice would gain the highest weight.If the mutation occurs in both the db genes, there would be no receptor to respond to the hormone produced. The mice would gain fat and produce the hormone in turn. And the hormone level would be abnormally high. Thus, lowest levels of hormone would be there in the mutated ob genes and highest levels of the hormone would be in the mutated db genes.

The hypothesis by Coleman was that the product of the ob+ gene was the appetite suppressing hormone.  Hence, the homozygous ob/ob mutant are in a position to synthesize that hormone, and its circulating level would be zero. He also hypothesized that the  product of the db+ gene was the receptor for the appetite-suppressing hormone. Thus, the homozygous db/db mutant would be able to synthesize the hormone but would not be in a position to respond to it. It would eat excessively and produce large amounts of body fat, which in turn would produce large amounts of appetite-suppressing hormone . In the absence of a receptor, the db/db mutant's hormone level would remain abnormally high. 
Coleman's hypothesis were confirmed when the precise functions of the ob+ and db+ genes were determined. The peptide hormone encoded by the ob+ gene was named Leptin.
