Why is saint of killers immune to genesis?

He isn't. In the comic book Preacher, Jesse managed to use genesis to stop the Saint from killing others by ordering him to not shoot. If he is immune to it in the show, then that has yet to be explained because in the comics it is a bit different. Jesse's use of genesis is the only time he was prevented from killing.
