HaeIII is not used to create a recombinant DNA; this is because it is a four-base cutter that produces a short fragment.

HaeIII is an endonuclease restriction enzyme that unlikely to be used to construct a recombinant DNA. This is because the HaeIII enzyme has only four base pairs. This would produce many short fragments of the plasmid. It is isolated from the bacteria named as <em>Haemophilus aegyptius bacteria. </em>Mostly restriction enzymes have a sequence of four to eight nucleotides. Other enzymes are different HaeIII enzyme because it is considerably very short.  

An individual can compare recombinant plasmid with the original one by comparing different restriction sites present for the particular enzyme by the process of gel electrophoresis. DNA is cut with the help of restriction enzyme that produces small fragment can run of gel electrophoresis. Restriction enzymes recognize and cleave specific DNA sequences generating either blunt or sticky ends.
