- Miscalculation in food reserves.

- Harsh weather

- Diseases.

Explanation:

The passengers of Mayflower miscalculated the duration of travel between England  to United States. As a result, they ran out of food faster than they predicted and some of their members died from hunger.

They also started their journey during winter. Bad quality of shelter on their ships made some people died from the harsh weather.

On top of that, they suffered an outbreak of scurvy, pneumonia, and tuberculosis in the middle of their journey. Since they're stucked in the middle of the sea, the patients unable to get proper treatment and died.
