Americans have seemingly always believed that their nation was blessed with a divine mission. what does this mean? how did that

notion relate to the concept of manifest destiny? does such a sense of mission persist today?

It means that they thought God wanted them to explore America. This is what the manifest destiny stems from. It does not really persist today.
