When a blood clot forms, activation of fibrin occurs which is a network of fibrous mesh that traps cells in order to prevent blood loss. Formation of fibrin allows catalytically inactive plasminogen to bind to the fibrin network.

Plasminogen contains fibrin-binding domains of plasminogen called kringle domains which recognize and bind lysine residues of fibrin. This process allows tPA whic is serine protease to catalyze the conversion of plasminogen into plasmin which will start the breaking down process of the clot
