The scientist that disproved the spontaneous generation theory was Louis Pasteur, who boiled a culture broth free of microorganisms and let it stand for a long period of time. No organisms grew from the flask, but when Pasteur opened it to the environment the flask soon became full of bacteria. That proved that there were microorganisms that were responsible for the growing of living things and disproved the theory of Spontaneous generation.

Proteins are molecules found in the cells of all living organisms. A protein is composed of monomers called amino acids linked together by peptide bonds. To analyze proteins, a technique called SDS-PAGE (sodium dodecyl sulfate polyacrylamide gel electrophoresis) is used. <u>This technique is used to separate proteins according to their electrophoretic mobility, depending on polypeptide chain length, molecular mass, post-translational modifications and other factors</u>. SDS (sodium dodecylsulfate) is an anionic surfactant compound that breaks non-covalent bonds in proteins, denatures them causing these protein molecules to lose their native conformation and provides the polypeptide with a negative charge that is proportional to the chain length (the number of amino acids) and, therefore, to the molecular mass of the protein. The electrostatic repulsion created by the binding of SDS to the protein is one of the causes of the protein losing its native conformation, thus eliminating the differences in conformation of the different proteins to be separated in the gel.

An electric current is applied to the gel where the proteins are seeded, causing the negatively charged proteins to migrate through the gel in the direction of the anode (positively charged region). Short proteins will move more easily and faster through the pores of the gel, while larger proteins will find it more difficult to do so. After a certain time, the proteins will have migrated according to their size (measured in kDa or kiloDaltons). After electrophoresis, the gel should be stained with Coomassie blue or silver stain, allowing visualization of the separated proteins. After staining, the different proteins will appear as distinct bands on the gel. In addition, the higher the amount of a protein, the more intense the color of its band will be.

Extract No. 1 shows three bands: 25, 30 and 35 kDa. <u>This means that it has three different proteins</u>, each with a different molecular weight. Extract number 2 shows two bands: 25 and 35 kDa. <u>It means that this sample has only 2 proteins. And being bands of higher intensity, we can say that there is more of each protein.</u>

Sample 1 then, has more variety of proteins but less quantity of each one, while sample 2 has less variety but more quantity of the two proteins present.

The preceding section reviewed the major metabolic reactions by which the cell obtains and stores energy in the form of ATP. This metabolic energy is then used to accomplish various tasks, including the synthesis of macromolecules and other cell constituents. Thus, energy derived from the breakdown of organic molecules (catabolism) is used to drive the synthesis of other required components of the cell. Most catabolic pathways involve the oxidation of organic molecules coupled to the generation of both energy (ATP) and reducing power (NADH). In contrast, biosynthetic (anabolic) pathways generally involve the use of both ATP and reducing power (usually in the form of NADPH) for the production of new organic compounds. One major biosynthetic pathway, the synthesis of carbohydrates from CO2 and H2O during the dark reactions of photosynthesis, was discussed in the preceding section. In addition to being obtained directly from food or generated by photosynthesis, glucose can be synthesized from other organic molecules. In animal cells, glucose synthesis (gluconeogenesis) usually starts with lactate (produced by anaerobic glycolysis), amino acids (derived from the breakdown of proteins), or glycerol (produced by the breakdown of lipids). Plants (but not animals) are also able to synthesize glucose from fatty acids—a process that is particularly important during the germination of seeds, when energy stored as fats must be converted to carbohydrates to support growth of the plant. In both animal and plant cells, simple sugars are polymerized and stored as polysaccharides.

Gluconeogenesis involves the conversion of pyruvate to glucose—essentially the reverse of glycolysis. However, as discussed earlier, the glycolytic conversion of glucose to pyruvate is an energy-yielding pathway, generating two molecules each of ATP and NADH. Although some reactions of glycolysis are readily reversible, others will proceed only in the direction of glucose breakdown, because they are associated with a large decrease in free energy. These energetically favorable reactions of glycolysis are bypassed during gluconeogenesis by other reactions (catalyzed by different enzymes) that are coupled to the expenditure of ATP and NADH in order to drive them in the direction of glucose synthesis. Overall, the generation of glucose from two molecules of pyruvate requires four molecules of ATP, two of GTP, and two of NADH. This process is considerably more costly than the simple reversal of glycolysis (which would require two molecules of ATP and two of NADH), illustrating the additional energy required to drive the pathway in the direction of biosynthesis.
