Pancreatic amylase's optimum pH is basic, NaHCO3 is a base so it will provide the optimum pH for the enzyme to catalyse the breakdown of starch.

In tube 1 there will not be the breakdown of starch as there simply isn't any enzyme to break it down.

In Tube 2, the enzyme pancreatic amylase will most probably become denatured (loss of shape), as HCl is acidic which causes it to lose shape causing a change in the active site and will never be able to catalyse the breakdown of starch.

The active site of an enzyme would form enzyme-substrate complex thus lowering the activation energy of the reaction and high rate of reaction is found.

Explanation:

The energy required to start the reaction is activation energy. In our system enzymes catalyses the reaction by lowering the activation energy. When the substrate binds to the active site or catalytic site of enzyme, an enzyme-  substrate complex is formed. This complex reduces the time required  for substrate changing into product. The enzyme substrate complex results in the conformational changes and contributes in lowering of activation energy.
