It only forms on proteins that are going to the lysosome. The mannose-6-phosphate receptor crosses the Golgi membrane and interacts with both the mannose-6-phosphate-containing protein on the luminal side of the membrane and coat proteins on the cytosolic side of the membrane.

Mannose-6-phosphate residues, which are detected by mannose-6-phosphate receptors in the trans-Golgi network and packaged into clathrin-coated vesicles, direct acid hydrolases to lysosomes.

These soluble lysosomal enzymes carry a distinctive identifier in the form of mannose 6-phosphate (M6P) groups, which are added only to their N-linked oligosaccharides as they transit through the lumen of the cis Golgi membrane.
